Introduction Ultraviolet and visible spectrometers have been in general use for the last 35 years and over this  period have become the most important analytical  instrument in the modern day laboratory.  In many applications other techniques could be  employed but none rival UV‐Visible spectrometry  for its simplicity versatility speed accuracy and for its simplicity, versatility, speed, accuracy and  cost‐effectiveness.

Electromagnetic (EM) Radiation is a form of energy  and we are constantly reminded of its presence via  our sense of sight and ability to feel radiant heat.  It may be considered in terms of a wave motion  b d d f where the wavelength, λ, is the distance between  two successive peaks and given in meter units.

λ The frequency, ν, is the number of peaks passing a  given point per second (Hertz units or per second).

These terms are related so that: c = νλ

where  c = 3.00 x 108 m/s and is the velocity of light  in a vacuum in a vacuum. The behavior of EM radiation depends on its wavelength.  Higher frequencies have shorter wavelengths, and lower  frequencies have longer wavelengths.  All EM radiation travels at a constant speed, the speed of  p , p 8 light (3 x 10 m/s), and is composed of photons.

The frequency and the wavelength of a particular  radiation are related by an equation [1] involving  the speed of light, c.  Combining [1] & [2] gives the equation: g[ ] [ ]g q E =hc/λ

Thus, higher the frequency or the shorter the wavelength of the photon, the more energy is associated with it.

Energy and Energy Levels  A molecule of any substance has an internal energy. Internal energy is the sum of :  1.electronic energy‐‐movement of electrons between electronic energy levels (or orbitals) in the molecule. 2.vibrational energy‐‐regular periodic stretching‐and‐contracting stretching and contracting and the bending of and the bending of bonds within the molecule.

The electronic energy levels of simple molecules are  widely separated. Usually only the absorption of a high energy photon,  that is one of very short wavelength, can excite a  molecule from one level to another. E2 (excited state)

UV‐Visible Spectroscopy UV‐Visible Spectrophotometry เป็ นเทคนิควิเคราะห์ที่อาศัยหลักการดูดกลืนแสงของสารในช่วง UV 200-400 nm (near UV) และ visible 400-800 nm ไม่นิยมใช้ ความยาวคลื่นที่ตํ่ากว่า 200 nm เนื่องจาก oxygen, nitrogen และความชื ้นในอากาศสามารถดูดกลืนแสงในความยาว คลื่นช่วงนี ้ได้ ถ้ถาเปนการวเคราะหสารมสหรอทาใหสเกดขน าเป็ นการวิเคราะห์สารมีสีหรื อทําให้ สีเกิดขึ ้น ซึซงดู ง่ ดดกลื ดกลนแสง นแสง ในช่วง visible จะเรี ยกวิธีการวิเคราะห์วา่ colorimetry ใช้ ทงในงานวิ ั้ เคราะห์เชิงปริมาณ (quantitive analysis) และเชิง คุณภาพ (quanlitative analysis)

การดูดกลืนแสง (Absorption) เกิดขึ ้นได้ เมื่อความแตกต่างของระดับพลังงานใน ground state และ excited state เท่ากับพลังงานของคลื่นแสงหรื อโฟตอนที่อะตอมหรื อโมเลกุลนัน้ ดูดดกลื ดกลนน เมื่อสารดูดกลืนแสงจะมีการถ่ายเทพลังงานจากแสงไปยังสารทําให้ อิเล็คตรอ นของสารนันเปลี ้ ่ยนระดับพลังงานจากระดับตํ่าหรื อสถานะพื ้น (ground state) ไปสูส่ ถานะกระตุ้น (excited state) ที่มีระดับพลังงานสูงกว่า อิเล็คตรอ นจะอยูใ่ นสถานะกระตุ้น 10-8-10-9 วินาที อิเล็คตรอนจะกลับลงสูส่ ถานะพื ้น โดยการคายแสงหรื อความร้ อนออกมา โดยการคายแสงหรอความรอนออกมา พลังงานคลื่นแสงที่ถกู ดูดกลืนจะมีคา่ เท่ากับผลต่างของระดับพลังงานของ groud state และ excited state

การดูดกลืนแสงของโมเลกุล เมื่อโมเลกุลของสารดูดกลืนแสงจะเกิดปรากฏการณ์ดงั ต่อไปนี ้ 1. 2. ช่วงของค่าการดูดกลืนแสง (A) และค่าการส่งผ่าน (T) จาก การวัดที่ให้ คา่ ความคลาดเคลื่อนสัมพัทธ์สงู เกินไป ควรวัดค่ าการดูดกลืนแสงให้ อยู่ในช่ วง A = 0.18-0.82 (หรื อ ≈0.1-1.0) หรื อ ค่ าการส่ งผ่ านในช่ วง 15-65 %T กําหนดให้

dA = ความคลาดเคลื่อนสัมบูรณ์ของ A ที่อ่านได้ จากเครื่ องมือ dT = ความคลาดเคลื่อนสัมบูรณ์ของ T ที่อ่านได้ จากเครื่ องมือ ดังนัน้ ค่าความคลาดเคลื่อนสัมพัทธ์ของ A และ T หาได้ จาก dA/A และ dT/T และความสัมพันธ์ระหว่างค่าความคลาดเคลื่อนสัมพัทธ์ของ C, A และ T หาได้ ดงั นี ้ dC/C = dA/A = ‐(1/lnT)(dT/T)

เมื่อสร้ างกราฟแสดงความสัมพันธ์ ระหว่ างค่ าความคลาดเคลื่อนสัมพัทธ์ ของความเข้ มข้ น (dC/C) และค่ าการดูดกลืนแสง(A)หรื อค่ าการส่ งผ่ าน (T) จะได้ ดังในรู ป
